    MsbA-dependent translocation of lipids across the inner membrane of Escherichia coli

    MsbA is an essential ABC transporter in Escherichia coli required for exporting newly synthesized lipids from the inner to the outer membrane. It remains uncertain whether or not MsbA catalyzes trans-bilayer lipid movement (i.e. flip-flop) within the inner membrane. We now show that newly synthesized lipid A accumulates on the cytoplasmic side of the inner membrane after shifting an E. coli msbA missense mutant to the non-permissive temperature. This conclusion is based on the selective inhibition of periplasmic, but not cytoplasmic, covalent modifications of lipid A that occur in polymysin-resistant strains of E. coli. The accessibility of newly synthesized phosphatidylethanolamine to membrane impermeable reagents, like 2,4,6-trinitrobenzene sulfonic acid, is also reduced severalfold. Our data showed that MsbA facilitates the rapid translocation of some lipids from the cytoplasmic to the periplasmic side of the inner membrane in living cells

    Learning and Wage Dynamics

    We develop a dynamic model of learning and wage determination: education may convey initial information about ability, but subsequent performance observations also are informative. Although the role of schooling in the labor market's inference process declines as performance observations accumulate, the estimated effect of schooling on the level of wages is independent of labor-market experience. In addition: time-invariant variables correlated with ability but unobserved by employers are increasingly correlated with wages as experience increases; wage residuals are a martingale; and wage cuts -are not rare, even for workers who do not change jobs. We present evidence from the National Longitudinal Survey of Youth that is generally consistent with all four of the model's predictions. We conclude that a blend of the learning model with an on-the-job-training model is more plausible than either model alone.

    Stochastic Inflation Revisited: Non-Slow Roll Statistics and DBI Inflation

    Stochastic inflation describes the global structure of the inflationary universe by modeling the super-Hubble dynamics as a system of matter fields coupled to gravity where the sub-Hubble field fluctuations induce a stochastic force into the equations of motion. The super-Hubble dynamics are ultralocal, allowing us to neglect spatial derivatives and treat each Hubble patch as a separate universe. This provides a natural framework in which to discuss probabilities on the space of solutions and initial conditions. In this article we derive an evolution equation for this probability for an arbitrary class of matter systems, including DBI and k-inflationary models, and discover equilibrium solutions that satisfy detailed balance. Our results are more general than those derived assuming slow roll or a quasi-de Sitter geometry, and so are directly applicable to models that do not satisfy the usual slow roll conditions. We discuss in general terms the conditions for eternal inflation to set in, and we give explicit numerical solutions of highly stochastic, quasi-stationary trajectories in the relativistic DBI regime. Finally, we show that the probability for stochastic/thermal tunneling can be significantly enhanced relative to the Hawking-Moss instanton result due to relativistic DBI effects.Comment: 38 pages, 2 figures. v3: minor revisions; version accepted into JCA

    Characterization of pPCP1 Plasmids in Yersinia pestis Strains Isolated from the Former Soviet Union

    Complete sequences of 9.5-kb pPCP1 plasmids in three Yersinia pestis strains from the former Soviet Union (FSU) were determined and compared with those of pPCP1 plasmids in three well-characterized, non-FSU Y. pestis strains (KIM, CO92, and 91001). Two of the FSU plasmids were from strains C2614 and C2944, isolated from plague foci in Russia, and one plasmid was from strain C790 from Kyrgyzstan. Sequence analyses identified four sequence types among the six plasmids. The pPCP1 plasmids in the FSU strains were most genetically related to the pPCP1 plasmid in the KIM strain and least related to the pPCP1 plasmid in Y. pestis 91001. The FSU strains generally had larger pPCP1 plasmid copy numbers compared to strain CO92. Expression of the plasmid's pla gene was significantly (P ≤ .05) higher in strain C2944 than in strain CO92. Given pla's role in Y. pestis virulence, this difference may have important implications for the strain's virulence

    The vacuum bubbles in de Sitter background and black hole pair creation

    We study the possible types of the nucleation of vacuum bubbles. We classify vacuum bubbles in de Sitter background and present some numerical solutions. The thin-wall approximation is employed to obtain the nucleation rate and the radius of vacuum bubbles. With careful analysis we confirm that Parke's formula is also applicable to the large true vacuum bubbles. The nucleation of the false vacuum bubble in de Sitter background is also evaluated. The tunneling process in the potential with degenerate vacua is analyzed as the limiting cases of the large true vacuum bubble and false vacuum bubble. Next, we consider the pair creation of black holes in the background of bubble solutions. We obtain static bubble wall solutions of junction equation with black hole pair. The masses of created black holes are uniquely determined by the cosmological constant and surface tension on the wall.     A Quintessentially Geometric Model

    We consider string inspired cosmology on a solitary D3D3-brane moving in the background of a ring of branes located on a circle of radius RR. The motion of the D3D3-brane transverse to the plane of the ring gives rise to a radion field which can be mapped to a massive non-BPS Born-Infeld type field with a cosh potential. For certain bounds of the brane tension we find an inflationary phase is possible, with the string scale relatively close to the Planck scale. The relevant perturbations and spectral indices are all well within the expected observational bounds. The evolution of the universe eventually comes to be dominated by dark energy, which we show is a late time attractor of the model. However we also find that the equation of state is time dependent, and will lead to late time Quintessence.Comment: 11 pages, 3 figures. References and comments adde
